干扰CD151基因表达对裸鼠宣威肺腺癌XWLC-05细胞肺转移的影响

Abstract:
摘要 目的 探讨RNA干扰抑制CD151基因表达对宣威肺腺癌XWLC-05细胞侵袭和迁移的影响及裸鼠肺转移的实验。方法 构建能高效抑制CD151基因表达的3组重组干扰质粒,筛选出稳定转染干扰质粒的宣威肺腺癌XWLC-05细胞株。MTT、划痕实验、Transwell实验研究干扰CD151基因表达前后对细胞迁移和侵袭的影响。将构建的RNA干扰抑制CD151基因表达的稳转细胞XWLC-05-shRNA、XWLC-05-shNC、XWLC-05-Blank通过尾静脉注射入BALB/c裸鼠,30天后通过肉眼和HE染色后观察肺转移情况。结果 划痕实验和Transwell实验表明RNA干扰抑制CD151基因表达可能抑制宣威肺腺癌XWLC-05细胞体外迁移和侵袭作用(P<0.05)。XWLC-05-shRNA、XWLC-05-shNC、XWLC-05-Blank各组裸鼠肺转移率及肺组织上的转移病灶数依次为75%(6/8)与(5.2±1.2)、100%(8/8)与(9.8±1.6)、87.5%(7/8)与(9.1±1.2)。XWLC-05-shNC与XWLC-05-Blank组比较差异无统计学意义(P=0.423),XWLC-05-shRNA与XWLC-05-shNC(P=0.000)和XWLC-05-Blank组比较差异有统计学意义(P=0.000)。结论 RNA干扰抑制CD151基因表达能抑制XWLC-05细胞的体外迁移和侵袭;能减少XWLC-05细胞裸鼠的肺转移。CD151可能成为抑制宣威肺癌侵袭和迁移的潜在治疗靶点之一
