西洋参多糖提取与纯化工艺优化

Abstract:
摘要 目的:筛选西洋参多糖的最佳提取及纯化工艺.方法:以西洋参多糖为指标,采用正交实验对料液比、煎煮时间与次数进行优化.以吸附率为指标,对上样液质量浓度、上样液pH、上样流速、径高比进行正交实验优选;以解吸率为指标,对乙醇体积分数、洗脱剂pH、洗脱流速采用正交实验进行优化.结果:料液比、提取次数是西洋参多糖提取的关键影响因素,西洋参多糖最佳提取工艺为料液比1∶10,加水提取3次,每次1.5 h.上样液质量浓度、上样液pH、上样流速、径高比为多糖纯化的关键影响因素,多糖的纯化工艺为上样质量浓度2.74 mg/mL,上样液pH 8.0,上样流速2.0 mL/min,径高比1∶8,最大上样量3.0 mL,洗脱剂为体积分数30%乙醇,洗脱剂pH 8.0,洗脱流速1.0 mL/min,洗脱剂用量3.0 mL;得到西洋参多糖纯度为47.94%.结论:建立的方法稳定、可靠,为开发利用西洋参资源提供基础.
