Ad-HIF-1α双突变体对大鼠缺血心肌及Na+-K+ATP酶的影响

Abstract:

摘要 目的:研究重组腺病毒低氧诱导因子-1α双突变体基因对心肌梗死大鼠缺血心肌损伤程度及Na+-K+ATP酶的影响.方法: 36只雄性SD大鼠建立急性心肌梗死模型,随机分成3组,分别为生理盐水组(Saline组,n=12)、腺病毒空载体组(Ad-Null组,n=12)、Ad-HIF-1α-564/402组(n=12).在术后即刻分别肌注生理盐水、Ad-Null、Ad-HIF-1α-564/402 0.1 mL,病毒滴度1.0×108 PFU.术后7 d,记录大鼠24 h饮食量、饮水量及体质量,观察大鼠的精神及运动状况.检测血清中肌酸激酶同工酶、肌钙蛋白I、B型利钠肽浓度.进行血流动力学检测,测量左室收缩压、左室舒张末压、左室最大上升/下降速率.采用分光光度计法检测心肌梗死大鼠心肌Na+-K+ATP酶水平.结果: 基因转染后7 d,3组大鼠饮食量、饮水量、体质量无统计学差异.Ad-HIF-1α-564/402组大鼠较Saline组、Ad-Null组反应更为灵敏,活动耐量增加.CK-Mb、cTnI、BNP血清浓度在Ad-HIF-1α 564/402组均较其他两组为低,3组间有统计学差异(P<0.05).Ad-HIF-1α-564/402组大鼠LVSP、±LVdp/dtmax明显高于Saline组、Ad-Null组(P<0.05).Ad-HIF-1α-564/402组大鼠LVEDP明显高于Saline组、Ad-Null组(P<0.05),上述指标Saline组、Ad-Null组均无统计学差异(P>0.05).分光光度计结果显示:Na+-K+ATP酶水平3组间无统计学差异(P>0.05).结论: Ad-HIF-1α-564/402能减轻心肌细胞梗死程度,保护心脏功能,对Na+-K+ATP酶水平无明显影响.
