Abstract:

摘要 目的: 构建一个基于博来霉素筛选的核糖体区基因打靶载体,为后续的基因打靶研究奠定基础.方法:首先选择含有博来霉素(zeocin)抗性的目的片段与载体连接,并进行单酶切及测序鉴定,最终构建基于博来霉素筛选核糖体基因打靶载体平台.结果:构成pUC19-DS1-SV40-Zeocin-polyA-DS2基因打靶载体与预期一致,测序结果与实验设计相同.结论:成功构建了含有抗性基因的pUC19-DS1-SV40-Zeocin-polyA-DS2水牛基因通用打靶载体平台,便于不同外源基因的插入,为转基因动物模型的建立提供了基础工具.
