Abstract:

调控蛋白质在凝胶中的扩散行为在生物医学领域具有重要的意义.本文采用巯基-烯点击反应设计并合成了一系列右旋糖酐基水凝胶.根据溶胀行为计算了凝胶的网格尺寸, 并以FITC标记的溶菌酶(FITC-Lyz)和牛血清白蛋白(FITC-BSA)为模型蛋白, 对蛋白质在水凝胶中的扩散行为进行了研究.结果表明, 以点击化学作为合成手段, 通过改变前驱体质量分数和烯/巯基摩尔比可以实现对凝胶网格尺寸的控制, 进而控制蛋白质在其中的扩散行为.
The regulation of protein diffusion in hydrogel is of great significance in biomedical field. In this study,a series of dextran-based hydrogels were designed and synthesized by applying “thiol-ene” click reaction. On the basis of swelling test result,the mesh size of hydrogel was calculated. Using FITC-labeled lysozyme(FITC-Lyz)and FITC-labeled bovine serum albumin(FITC-BSA)as model proteins,their diffusion behaviors in the hydrogel were investigated in detail. The results proved that the mesh size of hydrogel could be controlled by precursor mass fraction and ene/thiol molar ratio via click chemistry,leading to the tunable diffusion of proteins in hydrogel
