中药复方多糖对不同MHC B-Lβ II基因型蛋鸡免疫活性的影响

Abstract:
探究肌注中药复方多糖对不同MHC B-Lβ II基因型蛋鸡血清中细胞因子质量浓度(IL-1β、IL-10、IL-15)与IBDV抗体效价的影响规律。采用PCR-SSCP方法对试验鸡按不同基因型进行分组,将各组分成高、中、低剂量的多糖试验组和空白对照组。各组鸡分别于8日龄时肌肉注射0.2 mL的50 mg/mL、25 mg/mL、12.5 mg/mL中药复方多糖和生理盐水,连续7 d,于21、27、35、42和49日龄时从各组随机抽取5只鸡,翅下静脉采血并分离血清,利用ELISA法检测各试验组和空白组21、27、35、42和49日龄血清中细胞因子的质量浓度及IBDV抗体效价。结果显示,中药复方多糖组血清中IL-1β和IL-15的质量浓度及IBDV抗体效价均显著高于对照组,而IL-10的质量浓度则显著低于对照组,且不同剂量多糖组和基因型间存在差异。其中,AA基因型鸡中剂量多糖组的血清中IL-1β和IL-15的质量浓度及IBDV抗体效价显著高于高、低剂量组,IL-10显著低于高、低剂量组;BB基因型鸡高剂量多糖组的血清中IL-1β和IL-15的质量浓度及IBDV抗体效价显著高于中、低剂量组,IL-10显著低于中、低剂量组;BC基因型鸡中各剂量多糖组间血清中IL-1β、IL-15和IL-10的质量浓度及IBDV抗体效价并显著差异。结果表明,不同MHC B-Lβ II基因型鸡个体之间存在免疫应答能力的差异,不同剂量中药复方多糖对调节鸡免疫活性的最适基因型不同。
To investigate the effects of intramuscular injection of traditional Chinese medicinal compound polysaccharides on serum cytokines(IL-1β,IL-10,IL-15) and IBDV antibody titer in different MHC B-Lβ II genotypes of laying hens. PCR-SSCP technique was applied to group the experimental hens according to different MHC B-Lβ II genotypes,the hens were divided into four groups. Hens in each group at eight-day-age were intramuscularly injected with 0.2 mL traditional Chinese medicinal compound polysaccharides with the concentration of 50 mg/mL(high dose),25 mg/mL(medium dose),12.5 mg/mL(low dose) and physiological saline(control),continuously for 7 days. At 21-,27-,35-,42- and 49-day-old of ages,five chicken blood samples in each group were collected,and sera were separated for detection of cytokine contents and IBDV antibody titers. The results showed that,the mass concentrations of IL-1β,IL-15 and the titer of IBDV in the serum of compound polysaccharide groups were significantly higher than those in the control group,while the mass concentration of IL-10 was significantly lower than that of the control group,and there were differences between different doses of polysaccharides and genotypes. Among them,the mass concentrations of IL-1β,IL-15 and the titer of IBDV in serum of AA genotype chickens were significantly higher than those of high and low dose groups,while IL-10 was significantly lower than those of high and low dose groups.The mass concentrations of IL-1β,IL-15 and IBDV antibody in serum of BB genotype chickens of high dose polysaccharide group were significantly higher than those of middle and low dose group,while IL-10 was significantly lower than those of middle and low dose group.There were no significant differences between the concentration of IL-1β,IL-15,IL-10 and IBDV antibody in serum of BC genotype chickens.
