热激结合水杨酸处理对蝴蝶兰组培褐变的影响

Abstract:

以蝴蝶兰‘内山姑娘’品种的组培苗叶片为试材,将不同浓度的水杨酸(SA)添加于培养基中观察叶片的褐变情况,筛选出SA的适宜使用浓度为0.1 mmol·L-1。测定热激处理(40 ℃,9 min)和水杨酸处理(SA,0.1 mmol·L-1)单独使用以及协同作用时对叶片褐变指数、总酚(TP)质量分数、苯丙氨酸解氨酶(PAL)、多酚氧化酶(PPO)、过氧化物酶(POD)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和抗坏血酸过氧化物酶( APX)活性的影响。结果表明,热激、水杨酸处理以及热激结合水杨酸处理均能减轻蝴蝶兰叶片组培褐变,可降低蝴蝶兰叶片褐变过程中的褐变指数、总酚质量分数和PAL和PPO活性,提高SOD、CAT、APX活性和叶片诱导率。几种处理比较可见,热激与水杨酸协同作用抑制蝴蝶兰组培褐变的效果最好。
Phalaenopsis ‘Neyshanguniang’ variety was cultured on medium with different concentrations of salicylic acid (SA) to observe blade situation, and selected the best usage of SA concentration (0.1 mmol·L-1) and determined the activity of browning index, TP, PAL, PPO, POD, SOD, CAT and APX under the condition of heat-shock treatment (40 ℃, 9 min), salicylic acid (SA, 0.1 mmol·L-1) treatment and heat-shock combined with salicylic acid treatment, respectively. The results showed that heat-shock, salicylic acid and heat-shock combined with salicylic acid treatment could reduce browning index, total phenol content, PAL and PPO activity, increase the SOD, CAT and APX activity, and heat-shock combined with salicylic acid treatment has the best effect on reducing browning.
