Abstract:

利用反向遗传学方法,鉴定拟南芥(Arabidopsis thaliana)对大豆疫霉菌(Phytophthora sojae)非寄主抗病性相关的基因。通过分子生物学和生物信息学方法,鉴定和分析T-DNA插入拟南芥突变体离体叶片对大豆疫霉菌的非寄主抗病性。结果表明:4个果胶乙酰酯酶基因 AtPae7(pectin acetyl esterase 7)突变体株系中的3个对大豆疫霉菌的抗性减弱,表现为感病;qRT-PCR检测表明突变体中 AtPae7的转录水平下降;生物信息学预测PAE7蛋白的N端有23个氨基酸信号肽序列,确定其为胞外分泌蛋白。说明果胶乙酰酯酶基因 AtPae7参与拟南芥对大豆疫霉菌的非寄主抗病性。
This study is aimed at identification and characterization of genes related in non-host resistance in T-DNA insertional Arabidopsis thaliana mutants to Phytophthora sojae based on reversed genetics. A.thaliana T-DNA insertional mutants were identified and characterized with their resistance to P.sojae infecting detached leaves by molecular biological and bioinformatics methods. Three of four pae7 (pectin acetyl esterase 7) mutants presented a decrease of resistance to P.sojae and became susceptible. Transcriptional level of pae7 in its mutants declined based on detection by qRT-PCR. Bioinformatics analysis predicted that there were signal peptides of 23 amino acids at N-end of PAE7 protein, the results suggested that it is extracellular secreted protein. A.thaliana pae7 plays a role in non-host resistance of A.thaliana to P.sojae.
