Abstract:

从小麦条锈菌吸器cDNA文库中鉴定到一个泛肽-核糖体蛋白S27a(Ubiquitin-ribosomal protein S27a,UBRS27a)基因,命名为 PsUBRS27a。该基因开放阅读框为480 bp,编码包含159个氨基酸残基的蛋白质,其氨基酸序列含有典型的UBRS27a蛋白所具有的ubiquitin 和Ribosomal_S27结构域;序列比对发现UBRS27a蛋白在不同物种中高度保守,但PsUBRS27a蛋白在进化上与柄锈菌属的UBRS27a蛋白亲缘关系最近;种内多态性分析发现 PsUBRS27a基因在不同小麦条锈菌菌系中的序列非常保守,无核苷酸变化;实时荧光定量PCR 结果表明, PsUBRS27a基因在条锈菌侵染小麦过程中呈上调表达趋势,高峰期为接种后168 h。
We identified an ubiquitin-ribosomal protein S27a gene (designated PsUBRS27a) from the cDNA library of isolated haustoria of Puccinia striiformis f. sp.tritici(Pst). PsUBRS27a has an open reading frame of 480 bp that encodes a 159 aa protein whose N terminal and C terminal contain a ubiquitin domain and a ribosomal_S27 domain, respectively. Further sequence alignment revealed that PsUBRS27a proteins are highly conserved in different organisms, but phylogenetic analysis indicated that the PsUBRS27a protein is more closely related to UBRS27a proteins from Puccinia group. The intra-species polymorphism of PsUBRS27a was studied and the results indicated that PsUBRS27a was highly conserved in different Pst isolates and no single-nucleotide polymorphism was observed. qRT-PCR analysis showed that transcript level of PsUBRS27a was induced in planta during Pst infection and was peaked at 168 hour post-inoculation. This study provides important theoretical basis for further studying of the role of UBRS27a in Pst pathogenicity.
