利用EMS诱变构建柳枝稷穗芽无性系突变体库的初步研究

Abstract:
为探索创制柳枝稷(Panicum virgatum )无性系突变体库的适宜方法,以组织培养繁殖的柳枝稷品种‘Alamo’的穗芽作为外植体材料,利用甲基磺酸乙酯(EMS)对其进行诱变处理,并对EMS处理的最佳浓度和时间组合以及M1代田间生物学性状变异进行分析。结果表明:EMS诱变处理的最佳组合为用浓度0.047 mol/L的EMS处理4 h;M1代田间性状检测表明,变异群体在株高、分蘖数、开花期等性状上存在广泛变异,初步创制出一个柳枝稷穗芽无性系突变体库。
The aim of the study is to get a suitable method for creating mutant library for Panicum virgatum clones’ under the same genetic background,so as to provid valuable material for modern biotechnology breeding and molecular research in Panicum virgatum.We used multiple spike buds clumps of Panicum virgatum material ‘Alamo’ which obtained by tissue culture as explant material for EMS mutagenesis,the optimal EMS concentration,mutagenesis time and M1 generation biological character variation were analysised in the field.The results showed that the best mutagenic treatment for Panicum virgatum was 0.047 mol/L EMS concentration with 4 h,M1 generation field tests showed significant differences in plant height,tiller number per plant and blossom time.We preliminary created a mutant library with switchgrass spike buds in this study.
