miRNA-223-3p调控JAK2/STAT3信号通路对胃癌细胞增殖、凋亡、迁移的影响

Abstract:
目的:探讨miRNA-223-3p对胃癌细胞增殖、凋亡、迁移及JAK2/STAT3信号通路表达的影响。方法:RT-PCR检测40例胃癌组织及相应癌旁组织中miRNA-223-3p表达。培养人胃癌细胞株MKN-28,分别将miRNA-223-3p对照和miRNA-223-3p抑制物转染到细胞中,RT-PCR检测2组细胞中miRNA-223-3p的表达; CCK8法检测转染24、48、72 h后细胞的增殖情况; 流式细胞仪检测转染48 h后细胞凋亡率; Transwell小室法检测转染48 h后细胞的迁移数; Western blot 法检测Bcl-2、Bax、Cleaved-Caspase3、JAK2、STAT3、p-STAT3蛋白的表达情况。结果:miRNA-223-3p在胃癌组织中的表达高于癌旁组织(P=0.001); 转染miRNA-223-3p抑制物后miRNA-223-3p表达明显低于miRNA-223-3p对照组(P<0.001); miRNA-223-3p抑制组与miRNA-223-3p对照组比较,细胞增殖受到抑制,细胞凋亡率升高,细胞迁移数降低,Bax、Cleaved-Caspase3蛋白表达升高,JAK2、STAT3、p-STAT3、Bcl-2蛋白表达降低(P<0.05)。结论:miRNA-223-3p在胃癌组织中高表达,下调 miRNA-223-3p表达能抑制细胞增殖和迁移,促进细胞凋亡,其作用机制与JAK2/STAT3信号通路的调节有关。
Aim:To explore the effect of miRNA-223-3p by regulation of JAK2/STAT3 signaling pathway on proliferation, apoptosis and migration of gastric cancer cells.Methods:The expression of miRNA-223-3p in 40 cases of gastric cancer and adjacent tissue was detected by RT-PCR. Human gastric cancer MKN-28 cells were cultured,miRNA-223-3p control(miRNA-223-3p control group)and miRNA-223-3p inhibitor(miRNA-223-3p inhibitor group)was transfected into cells, and the expression of miRNA-223-3p was detected by RT-PCR. CCK8 assay was used to detect cell proliferation after 24, 48, 72 h. Cell apoptosis was detected after 48 h by flow cytometry. Cell migration was detected after 48 h by Transwell assay. The expressions of Bcl-2, Bax, Cleaved-Caspase3, JAK2, STAT3, p-STAT3 protein were detected by Western blot.Results:The expression of miRNA-223-3p in gastric cancer tissue was significantly higher than that of para cancerous tissue(P=0.001), and the expressions of miRNA-223-3p was significantly lower than that of the miRNA-223-3p control group after transfection of miRNA-223-3p inhibitor(P<0.001). Compared with the miRNA-223-3p control group, the proliferation of cells decreased in miRNA-223-3p inhibitor group, the apoptosis rate increased, the number of migration cells decreased, the expressions of Bax and Cleaved-Caspase3 protein increased, and the expressions of JAK2, STAT3, p-STAT3 and Bcl-2 protein decreased(P<0.05).Conclusion:miRNA-223-3p is highly expressed in gastric cancer, down-regulation of miRNA-223-3p could inhibit cell proliferation and migration, promote cell apoptosis, and its mechanism is related to the regulation of JAK2/STAT3 signaling pathway
