Abstract:

目的:探讨光照强度和盐浓度对杜氏盐藻生长和细胞内β?埠?萝卜素积累的影响。方法:检测正常培养条件下杜氏盐藻生长情况和β?埠?萝卜素变化,不同浓度(0.5、1.5、2.0、2.5、3.0、3.5 mol/L)NaCl培养后细胞内β?埠?萝卜素的积累情况。采用高光(5 000~6 000 Lux)、高盐(3.5 mol/L NaCl)及高光高盐条件对培养至对数生长末期的杜氏盐藻进行β?埠?萝卜素积累的诱导。结果:正常条件下杜氏盐藻培养2 周后进入稳定期,细胞内β?埠?萝卜素含量呈先下降后上升趋势,第9 天达到最大值,随后出现小幅度波动;杜氏盐藻细胞内β?埠?萝卜素含量随NaCl浓度升高而升高,二者呈正相关(r=0.969,P<0.001)。与正常条件相比,高光、高盐单独作用均能够提高细胞内β?埠?萝卜素含量,而高光高盐联合作用最有利于杜氏盐藻细胞β?埠?萝卜素的积累。结论:高光高盐联合作用可作为杜氏盐藻细胞内β?埠?萝卜素的最佳诱导条件。
Aim: To study the effects of light and salt concentration on the growth of Dunaliella salina and intracellular accumulation of β??carotene. Methods: The growth of Dunaliella salina and change of β??carotene content were measured under normal culture conditions. And the accumulation of β??carotene under different salt concentrations(0.5,1.5,2.0,2.5,3.0,3.5 mol/L) was detected. Then high??light(5 000-6 000 Lux), high??salt(3.5 mol/L NaCl) and high??light plus high??salt conditions were used to induce β??carotene accumulation. Results: Under normal conditions, Dunaliella salina grew to the stable phase after 2 weeks of cultivation,the intracellular β??carotene content was decreased at first and then increased,on the 9th day the content reached a maximum followed by a slight fluctuation. The β??carotene content of cells increased with increasing salt concentration, indicating that the two indexes were positively correlated(r=0.969,P<0.001). Furthermore, the results of induction experiments showed that the β??carotene content increased in both high??light and high??salt conditions, compared with normal conditions. And the combination of high??salt and high??light was most favorable to β??carotene accumulation. Conclusion: The combined high??light and high??salt might be the optimal induction conditions for β??carotene accumulation in Dunaliella salina,which provides a theoretical basis for the industrial production of Dunaliella salina
