卵泡液中游离DNA含量与卵母细胞成熟度及胚胎质量的关系

Abstract:
目的:探讨卵泡液中游离DNA含量与卵母细胞成熟度及胚胎质量的关系。方法:共收集了87位患者的成熟与未成熟卵泡液。以年龄30岁为界,定义年龄<30岁的为A组,年龄≥30岁的为B组。使用DNA手工提取法(改良酚-氯仿-异丙醇法)进行卵泡液中游离DNA的提取并检测。结果:成熟卵泡液组中游离DNA含量[(1 168.4±81.1)μg/L]低于未成熟卵泡液组[(1 875.6±95.7)μg/L],差异有统计学意义(P<0.001)。A组可移植胚胎数目[(5.6±2.7)个]、优质胚胎数目[(3.9±1.8)个]高于B组[(4.1±2.5)和(2.5±1.6)个],A组成熟卵泡液中游离DNA含量[(1 129.4±78.6)μg/L]低于B组[(1 214.2±56.8)μg/L],差异均有统计学意义(P<0.001)。结论:游离DNA含量可能是评价卵泡细胞成熟度以及预测胚胎质量的指标。
Aim: To explore the relationship between the content of cell-free DNA and oocyte maturation and embryo quality in follicular fluid.Methods: The mature and immature follicular fluid of 87 patients were collected. Patients with age<30 years were group A(n=47), and patients with age≥30 years were group B(n=40). The concentration of cell-free DNA was detected by manual extraction(modified phenol-chloroform-isoamyl alcohol extraction).Results: The content of cell-free DNA in the mature follicular fluid group[(1 168.4±81.1)μg/L)] was significantly lower than that in immature follicular fluid group [(1 875.6±95.7)μg/L)](P<0.001). The patients' partial parameters such as the number of embryo numbers(5.6±2.7), good quality embryo number(3.9±1.8)in group A were significantly higher than the patients in group B[(4.1±2.5)and(2.5±1.6)]. But the content of cell-free DNA in group A [(1 129.4±78.6)μg/L)] was lower than that in group B[(1 214.2±56.8)μg/L)](P<0.001).Conclusion: The content of cell-free DNA can be used as a biological indicator to evaluate oocyte maturation, and it may also be an indicator to predict the quality of embryos
