Abstract:

目的:观察H2O2对人小梁细胞(HTCs)内质网应激相关蛋白GRP78、CHOP及其mRNA表达的影响。方法:实验分为对照组、H2O2 6 h组、H2O2 12 h组和H2O2 24 h组,分别以600 μmol/L H2O2作用于HTCs 0、6、12及24 h后,采用Real-time PCR和Western blot检测细胞中CHOP、GRP78 mRNA和蛋白的表达。结果:对照组无GRP78和CHOP的表达; H2O2处理后HTCs GRP78、CHOP mRNA及蛋白的表达水平均升高,且随着H2O2作用时间的延长,GRP78、CHOP mRNA及蛋白的表达水平升高(P均<0.05)。结论:H2O2可诱导HTCs发生内质网应激,从而参与HTCs的氧化损伤。
Aim: To observe the effects of H2O2 on expressions of endoplasmic reticulum stress(ERS)markers GRP78 and CHOP in human trabecular meshwork cells(HTCs),so as to explore the role of ERS in the oxidative damage of HTCs.Methods: The experiment included control group, H2O2 6 h group, H2O2 12 h group and H2O2 24 h group, and HTCs were treated with 600 μmol/L H2O2 for 0,6,12 and 24 h,respectively. The expressions of GRP78 and CHOP mRNA and protein were determined by Real-time PCR and Western blot, respectively.Results: Compared with control group, the expression levels of GRP78 and CHOP mRNA and protein in various H2O2 treatment groups increased; the expressions levels of GRP78 and CHOP mRNA and protein tended to increase along with the prolongation of H2O2 treatment(P<0.05).Conclusion: Exogenous H2O2 could induce ERS in HTCs,which is involved in the oxidative damage of HTCs
