番泻叶水提物对3T3-L1前脂肪细胞增殖、凋亡及脂联素基因表达的影响*

Abstract:
目的:观察番泻叶水提物(FSAE)对3T3-L1前脂肪细胞增殖及凋亡影响,并进一步研究FSAE对3T3-L1前脂肪细胞脂联素mRNA及脂联素启动子荧光素酶活性的影响。方法:体外培养3T3-L1前脂肪细胞,给予不同质量浓度FSAE刺激24 h,MTT法检测细胞增殖,AnnexinV-FITC/PI双染法检测细胞凋亡,Western blot法检测细胞凋亡蛋白Caspase-3、Bax的表达,荧光定量PCR、双荧光素酶报告基因法检测FSAE对3T3-L1脂肪细胞脂联素mRNA和脂联素基因启动子活性的影响。结果:不同质量浓度FSAE对3T3-L1前脂肪细胞的增殖有抑制作用,能够促进细胞凋亡,提高凋亡蛋白Caspase-3、Bax蛋白的表达; 同时,FSAE能够增加3T3-L1前脂肪细胞中脂联素mRNA的表达,提高其启动子活性。结论:上述研究结果可能为FSAE抑制肥胖提供分子生物学依据。
Aim: To determine the effect of Folium Sennae aqueous extract(FSAE)on 3T3-L1 cell proliferation and apoptosis, and further study its effect on 3T3-L1 adiponectin mRNA and adiponectin promoter luciferase activity.Methods: 3T3-L1 cells were incubated with different concentrations of FSAE for 24 h, and cell proliferation was determined by MTT method; AnnexinV-FITC/PI double staining was used to detect cells apoptosis; Western blot was used to detect the expressions of Caspase-3 and Bax protein; Fluorescence quantitative PCR was used to detect adiponectin mRNA expression; double fluorescein enzyme report gene method was used to detect the influence of adiponectin gene promoter activity.Results: Different concentration FSAE inhibited 3T3-L1 proliferation,promoted cell apoptosis, improved the expressions of apoptosis related protein Caspase-3 and Bax; At the same time, FSAE improved adiponectin mRNA expression, and enhanced its adiponectin promoter activity.Conclusion: This may provide a molecular basis for FASE inhibiting obesity
