丹蒌片对同型半胱氨酸诱导的兔主动脉内皮细胞CHOP、BIP蛋白表达和凋亡的影响

Abstract:
目的:观察丹蒌片对同型半胱氨酸(HCY)诱导的兔主动脉内皮细胞凋亡及内质网应激相关蛋白C/EBP 环磷酸腺苷反应元件结合转录因子同源蛋白(CHOP)、免疫球蛋白重链结合蛋白(BIP)表达的影响。方法:以 HCY诱导建立兔主动脉内皮细胞损伤模型,分别用瑞舒伐他汀(他汀)、丹蒌片含药血清或二者联合共同培养48 h后,Annexin V-PI法检测细胞凋亡率,Western blot法检测CHOP、BIP蛋白表达情况。以未诱导细胞作空白对照。结果:模型细胞凋亡率及BIP、CHOP蛋白表达水平均高于空白对照(P<0.05)。丹蒌片和他汀可明显降低模型细胞的凋亡率,二者具有协同作用(F丹蒌片=475.288,F他汀=235.248,F交互=121.377,P<0.05); 丹蒌片可显著下调模型细胞BIP、CHOP蛋白的表达水平,与他汀无协同作用(F丹蒌片=7.017,8.147,P均<0.05; F交互=0.590,1.406,P均>0.05)。结论:丹蒌片可通过下调CHOP、BIP的表达抑制血管内皮细胞凋亡。
Aim: To study the effect of Danlou tablets on the apoptosis of rabbit aortic endothelial cells induced by homocysteine(HCY)and the expressions of endoplasmic reticulum stress related proteins C/EBP-homologous protein(CHOP)and immunoglobulin heavy chains binding protein(BIP).Methods: There were 5 groups. The blank group: aortic endothelial cells were not induced by HCY; the model group: aortic endothelial cells were induced by HCY; Danlou group, rosuvastatin group and combination group: aortic endothelial cells were induced by HCY and then cultured for 48 hours by serum containing Danlou tablet, rosuvastatin or rosuvastatin and Danlou tablet, respectively. Apoptosis rate and expressions of CHOP and BIP protein were detected by Annexin V-PI and Western blot.Results: Compared with blank group, apoptosis rate, the expressions of CHOP and BIP protein in model group increased significantly(P<0.05). Both Danlou tablet and rosuvastatin could decrease apoptosis rate of cells with HCY induction, and there was synergistic effect between them(FDanlou tablets=475.288,Frosuvastatin=235.248,Finteraction=121.377,P<0.05). Danlou tablet could decrease the expressions of BIP and CHOP protein(FDanlou tablets=7.017,8.147, P<0.05), but had no synergy with rosuvastatin(Finteraction=0.590,1.406,P>0.05).Conclusion: Danlou tablet could inhibit the apoptosis of vascular endothelial cells by down-regulating the expressions of CHOP and BIP protein
