黑米花色苷对哮喘小鼠气道炎症的影响

Abstract:
目的:观察黑米花色苷(AEBR)的抗哮喘作用并探讨其可能的作用机制。方法:50只雄性BALB/c小鼠随机分成正常组、模型组、AEBR低剂量组、AEBR中剂量组、AEBR高剂量组。体外卵清蛋白(OVA)诱导建立哮喘模型,支气管肺泡灌洗液(BALF)内细胞Diff-quik染色后观察细胞总数和各分类细胞数。肺组织切片HE染色观察肺部炎症情况。ELISA和Western blot方法分别检测BALF和肺组织中白细胞介素-4(IL-4)、IL-5、干扰素-γ(IFN-γ)水平的变化。Western blot检测细胞质和细胞核中NF-κB p65的变化。结果:与正常组相比,模型组炎症细胞计数、IL-4、IL-5和NF-κB p65核转位增高(P<0.05),IFN-γ表达降低(P<0.05); AEBR中、高剂量组炎症细胞数、IL-4和IL-5的表达以及NF-κB p65核转位明显降低(P<0.05),IFN-γ的表达升高(P<0.05)。结论:AEBR通过抑制NF-κB活化直接或间接影响Th1/Th2细胞因子失平衡而发挥抗哮喘作用。
Aim: To investigate the anti-asthma effects and possible mechanism of anthocyanin-rich from black rice(AEBR).Methods: Fifty male BALB/c mice were randomly allocated into five groups:control group,model group and AEBR groups of low,middle and high dose.Ovalbumin(OVA)was injected intraperitoneally and inhaled to produce the asthmatic model. The number of total cells and classified cells in bronchoalveolar lavage fluid(BALF)were observed by diff-quik staining. The lung tissue inflammation was observed by HE staining. The concentrations of IL-4, IL-5 and IFN-γ in BALF and lung tissue respectively measured by ELISA and Western blot. The changes of NF-κB p65 in cytoplasm and nucleus were analyzed by Western blot.Results: Compared with control group,the number of inflammatory cells, the concentrations of IL-4 and IL-5 and the NF-κB p65 nuclear translocation were enhanced,but IFN-γ were reduced in the model group(P<0.05); compared with the model group, the number of inflammatory cells, the concentrations of IL-4,IL-5 and NF-κB p65 nuclear translocation were decreased distinctly,while IFN-γ obviously increased in AEBR groups of middle and high dose(P<0.05).Conclusion: AEBR may inhibit NF-κB transcriptional activation and directly or indirectly regulate imbalance between Th1/Th2 cytokine to attenuate allergic airway inflammation
