Abstract:

目的:探讨鹿血晶对荷S180肉瘤小鼠放疗后免疫功能的影响及其可能机制,为临床用药提供依据。方法:建立S180荷瘤小鼠模型,分为模型组、放疗组、放疗+鹿血晶组,另设正常组,每组10只。放疗组、放疗+鹿血晶组小鼠给予X线局部照射,2 Gy/次,1次/d,连续5 d,放疗+鹿血晶组自照射开始即灌胃给予鹿血晶混悬液,连续给药10 d。治疗结束采用流式细胞术检测小鼠外周血T淋巴细胞亚群比例; ELISA法检测小鼠血清IFN-γ、IL-1、IL-2、IL-4、IL-6水平; 免疫组化法检测脾脏组织转录因子GATA-3和T-bet的表达。结果:与模型组比,放疗组荷瘤小鼠CD4+T淋巴细胞数量减少,CD4+/CD8+细胞比例降低,血清IFN-γ水平降低,IL-4水平升高,脾脏组织GATA-3、T-bet的表达均降低; 与放疗组比较,放疗+鹿血晶组荷瘤小鼠CD4+T淋巴细胞数及CD4+/CD8+细胞比例均增加,脾脏指数升高,血清IFN-γ升高,IL-1、IL-4水平下降,脾脏组织GATA-3的表达降低,T-bet表达的下降减缓(P均<0.05)。结论:鹿血晶对放疗荷瘤小鼠的免疫功能具有一定的保护作用,其可调节Th1/Th2 型细胞因子的比例,部分机制可能与其影响T淋巴细胞转录因子T-bet与GATA-3的表达有关。
Aim: To study the effects of cervus blood crystal on the immune function in S180 tumor-bearing mice after radiotherapy(RT)and its possible mechanism.Methods: Forty mice were allocated into 4 groups: normal group, model group, RT group, RT+cervus blood crystal group. The mice in the latter 3 groups were constructed S180 tumor-bearing mice model. The mice in RT group and RT+cervus blood crystal group were exposed to X-ray local irradiation, 2 Gy for each time, once per day for 5 days. Since the beginning of the radiation, the mice in RT+cervus blood crystal group were given cervus blood crystal suspension by gavage for 10 days. The levels of IFN-γ, IL-1, IL-2, IL-4 and IL-6 in serum of mice were detected by ELISA. The expressions of transcription factors GATA-3 and T-bet in spleen tissue were detected by immunohistochemistry. Results: After being given cervus blood crystal, the CD4+ T cells and the ratio of CD4+/CD8+ in peripheral blood of RT group all increased,the spleen index increased,the level of serum IFN-γ increased,while the levels of IL-1 and IL-4 decreased(P<0.05).Besides, cervus blood crystal also could reduce the expression of GATA-3,and slow down the decrease of T-bet(P<0.05).Conclusion: Cervus blood crystal has protective effects on the immune function of tumor-bearing mice undergoing RT. It can also regulate Th1/Th2 type cytokines, which may result from its impact on T-bet and GATA-3
