缺氧下膀胱癌细胞黏附能力和侵袭能力的变化*

Abstract:
目的:探讨缺氧条件下不同分化膀胱癌细胞5637和T24生物学特性变化。方法:将不同分化程度的膀胱癌细胞5637和T24分别置于正常氧浓度条件下(37 ℃, 体积分数5% CO2和体积分数21% O2)和缺氧(37 ℃、体积分数10%O2、体积分数5% CO2和体积分数85%N2)环境中培养,以MTT法和Boyden侵袭小室法检测两种膀胱癌细胞在不同环境下黏附能力和侵袭能力的差异。结果:随着培养时间的延长,缺氧组和对照组5637和T24细胞的黏附率逐渐增加,差异有统计学意义(P<0.001); 5637和T24细胞黏附率在缺氧24 h与对照组相比差异无统计学意义(P>0.05),但缺氧48 h和72 h两种细胞的黏附能力均高于对照组(P<0.05); 缺氧组5637和T24细胞的穿膜细胞数均高于对照组(P<0.001)。结论:缺氧条件下膀胱癌细胞的黏附和迁移能力增强。
Aim: To study the effects of hypoxia on adhesion and migration of bladder carcinoma cell line 5637 and T24 in vitro.Methods: The bladder carcinoma cells 5637 and T24 were cultured in hypoxic condition(10%O2,5% CO2,85%N2 mixed gas), while those put into 5% CO2,37 ℃ incubator were control group. The influence of hypoxia on the adhesiveness and invasiveness of the 2 kinds of cells were detected by MTT and Boyden method, respectively.Results: The result showed that the adhesiveness ability of 5637 cells and T24 cells was enhanced with culture time in hypoxic condition and the normal oxygen condition(P<0.001). Compared with those cultured in normal oxygen condition, the adhensiveness of the 2 cells cultured in hypoxic condition did not change significantly after 24 h culture(P>0.05), but increased after 48 or 72 h culture(P<0.05).The migration ability of the 2 kinds of cells was enhanced after cultured in hypoxic condition than that of control group(P<0.001).Conclusion: The adhesiveness and migration ability of bladder carcinoma cells are enhanced in hypoxic condition
