上皮性卵巢癌组织中Slug和E-cadherin的表达
Expressions of Slug and E-cadherin in epithelial ovarian cancer

目的:探讨Slug及E-cadherin在上皮性卵巢癌组织中的表达及其相关性。方法:分别采用免疫组化SP法和荧光定量PCR法检测上皮性卵巢癌组织(n=54)、卵巢良性上皮性肿瘤组织(n=30)、正常卵巢组织(n=20)中Slug和E-cadherin蛋白及mRNA的表达,分析2个因子蛋白阳性表达率与上皮性卵巢癌患者临床病理特征的关系和mRNA表达的相关性。结果:Slug蛋白在上皮性卵巢癌组织、卵巢良性上皮性肿瘤组织及正常卵巢组织中的蛋白阳性表达率分别为88.9%、43.3%和5.0%(P<0.001),E-cadherin蛋白分别为22.2%、46.7%和85.0%,差异有统计学意义(P<0.001)。上皮性卵巢癌组织中Slug和E-cadherin的蛋白阳性表达率在不同临床分期、有无淋巴结转移及有无腹水中的差异有统计学意义(P<0.05)。 在上皮性卵巢癌组织中Slug和E-cadherin mRNA的表达呈负相关(r=-0.276,P=0.044)。结论:Slug和E-cadherin在上皮性卵巢癌的发生、发展中发挥重要作用,可能与癌细胞的侵袭和转移也有关。
Aim: To investigate the expressions of Slug and E-cadherin in epithelial ovarian cancer tissue and their correlation.Methods: Immunohistochemical SP and RT-PCR method were used to examine the protein and mRNA expressions of Slug and E-cadherin in epithelial ovarian carcinoma tissue(n=54), ovarian benign epithelial tumor tissue(n=30)and normal ovarian tissue(n=20). The relationship between the protein positive expression rate of the 2 factors and clinicopathologic features in epithelial ovarian cancer and the correlation of mRNA expression of the 2 factors were analyzed.Results: The protein positive expression rates of Slug and E-cadherin in epithelial ovarian carcinoma tissue, benign epithelial ovarian tumor tissue and normal ovarian tissue were 88.9%,43.3%,5.0% and 22.2%,46.7%,85.0%, and the difference were statistically significant(P<0.001). The protein positive expression rates of Slug and E-cadherin in epithelial ovarian cancer tissue were statistically different in different clinical stages, with or without lymphatic metastasis and with or without ascites(P<0.05). The mRNA expression level of Slug was negatively correlated with E-cadherin in epithelial ovarian cancer tissue(r=-0.276,P=0.044).Conclusion: Slug and E-cadherin may play important roles in the occurrence and development of epithelial ovarian cancer and may involved in the invasion and metastasis of cancer cells
