姜黄素对高尿酸血症大鼠肾组织中TGF-β1、NF-κB表达的影响

Abstract:
目的:观察姜黄素对高尿酸血症(HUA)大鼠肾组织中转化生长因子-β1(TGF-β1)、核因子-κB(NF-κB)表达的影响。方法:SD大鼠36只,随机分为正常对照组、氧嗪酸钾组和氧嗪酸钾+姜黄素组(联合组),每组12只,氧嗪酸钾灌胃制备HUA模型,模型成功后联合组给予姜黄素灌胃治疗8周。检测大鼠血尿酸(UA)、肌酐(Scr)、尿素氮(BUN)和24 h尿蛋白定量及血清胱抑制素C(Cys-C)水平; HE染色及Masson染色观察肾脏病理变化情况,免疫组织化学法检测肾组织中TGF-β1、NF-κB蛋白的表达。结果:与对照组比较,氧嗪酸钾组及联合组UA、Scr、BUN、24 h尿蛋白定量、Cys-C水平均升高(P<0.05); 联合组较氧嗪酸钾组血清BUN、Scr、24 h尿蛋白定量、Cys-C均下降(P<0.05); HE染色显示氧嗪酸钾组肾小管炎性细胞浸润、肾小管轻度扩张,联合组较氧嗪酸钾组肾小管损伤程度减轻; Masson染色显示氧嗪酸钾组肾小管间质纤维化程度大于对照组及联合组,联合组纤维化程度减轻; 免疫组化检测结果显示氧嗪酸钾组TGF-β1 、NF-κB表达水平升高(P<0.05),联合组TGF-β1 、NF-κB表达量较氧嗪酸钾组下降(P<0.05)。结论:姜黄素对HUA肾脏具有保护作用,其机制可能与下调TGF-β1、NF-κB的表达有关。
Aim: To observe the effect of curcumin on the expressions of TGF-β1 and NF-κB in renal tissue of rats with hyperuricemia(HUA).Methods: Thirty-six male SD rats were randomly allocated into three groups: control group(NC)group, potassium oxonate(OP)group, and potassium oxonate with curcumin group(OC)group, with 12 rats in each group. Sodium carboxymethy cellulose solution at 5 g/L containing OP was used to establish hyperuricemia model by gastric gavage once per day. OC group was treated with curcumin at the same time.NC group was treated with 10 g/L sodium carboxymethy cellulose only.The levels of serum uric acid(UA), creatinine(Scr), urea nitrogen(BUN), 24-h urinary protein and cystatin C(Cys-C)were measured at 12th week.The histological changes of kidneys were observed via HE staining and Masson staining by light microscopy.The expressions of TGF-β1 and NF-κB in renal tissue were examined by immunohistochemistry staining.Results: Compared with NC group, the values of UA, Scr, BUN, 24-h urinary protein and Cys-C were higher in OP group and OC group(P<0.05); compared with OP group, the values of BUN,Scr, 24-h urinary protein and Cys-C in the OC group were lower at 12th week(P<0.05). HE and Masson staining showed that OC group had less damage and tubulointestitial fibrosis compared with OP group. The expressions of TGF-β1 and NF-κB in the glomeruli and renal tubules of the OP group were significantly higher than those in the control group(P<0.05). The expression of NF-κB,TGF-β1 was significantly down-regulated in OC group than that in OP group(P<0.05).Conclusion: The expressions of TGF-β1 and NF-κB in renal tissue of rats with HUA was up-regulated, and curcumin can protect the kidney by down-regulating the expressions of TGF-β1 and NF-κB in HUA
