四种人子宫内膜癌细胞系中IGFBP-rP1的定位及表达

Abstract:
目的:检测不同子宫内膜癌细胞系中胰岛素样生长因子结合蛋白相关蛋白1(IGFBP-rP1)的定位及表达情况。方法:体外培养子宫内膜癌细胞Ishikawa、RL95-2、HEC-1B和HEC-1A,细胞免疫荧光法检测IGFBP-rP1在4种子宫内膜癌细胞中的定位; 实时荧光定量PCR法与Western blot法检测4种子宫内膜癌细胞中IGFBP-rP1 mRNA及蛋白的表达水平。结果:细胞免疫荧光染色示IGFBP-rP1主要定位于4种子宫内膜癌细胞的胞浆。子宫内膜癌Ishikawa、RL95-2细胞中IGFBP-rP1 mRNA及蛋白表达水平明显高于HEC-1A、HEC-1B细胞(P<0.001)。HEC-1A 细胞中IGFBP-rP1 mRNA及蛋白表达量最低,Ishikawa细胞中表达量最高(P均<0.05)。结论:4种子宫内膜癌细胞系中均表达IGFBP-rP1。
Aim: To detect the expression of insulin-like growth factor binding protein-related protein 1(IGFBP-rP1)in different human endometrial carcinoma cell lines.Methods: The human endometrial carcinoma cell lines Ishikawa,RL95-2,HEC-1B and HEC-1A were cultured in vitro. The localization of IGFBP-rP1 in 4 different endometrial carcinoma cell lines was detected by cell immunofluorescence. Using fluorescence quantitative PCR and Western blot, we examined the mRNA and protein expression of IGFBP-rP1 in 4 endometrial carcinoma cell lines.Results: Immunofluorescence staining showed that IGFBP-rP1 was localized in the cytoplasm of 4 endometrial cancer cell lines. The expression of IGFBP-rP1 mRNA and protein in Ishikawa and RL95-2 cells were significantly higher than that in HEC-1A and HEC-1B cells(P<0.001). IGFBP-rP1 mRNA and protein had the lowest expression in HEC-1A cells and the highest expression in Ishikawa cells(P<0.05).Conclusion: IGFBP-rP1 is expressed in 4 human endometrial carcinoma cell lines
