Abstract:

目的:探讨Bcl-2、Bax 和Caspase-9在性成熟期小鼠动情周期不同阶段子宫内膜组织中的表达及定位。方法:选取32只性成熟期健康未孕雌性昆明小鼠,通过阴道脱落细胞检测法,根据阴道涂片细胞种类,确定各小鼠所属动情周期阶段,将小鼠分4组(n=8),分别为动情前期组、动情期组、动情后期组、动情间期组。免疫组化法检测Bcl-2、Bax和Caspase-9蛋白在子宫内膜组织中的表达。结果:Bcl-2在子宫内膜上皮组织中表现出与动情周期相关的规律性表达,动情间期表达高于其他各期(P<0.05); Bax在各动情周期均有表达,不同动情周期子宫内膜组织中Bax蛋白的表达差异无统计学意义(P=0.796); Caspase-9在动情期子宫内膜组织中主要表达于黏膜上皮细胞胞质、固有层子宫腺细胞胞质、基质细胞胞质,且动情前期表达低于其他各期(P<0.05)。结论:Bcl-2、Bax、Caspase-9在动情周期中共同调节子宫内膜细胞周期性的增殖与凋亡。
Aim: To explore the expressions of Bcl-2,Bax,and Caspase-9 in endometrium at different stages of estrous cycle in mature mice.Methods: A total of 32 sexual maturity healthy mice were randomly allocated into 4 groups(n=8),and through the rats vaginal smear observation and types of the cells, the mouse's estrous cycle stage was clearly determined.Then immunohistochemistry was carried out to screen the protein of Bcl-2,Bax,and Caspase-9 in mice.Results: Bcl-2 was highly appeared in diestrus stage(P<0.05).Bax was also expressed in the estrous cycle but had no significant difference among different stages(P=0.796).Besides, Caspase-9 in each estrus stage of mainly expressed in the mucosa epithelial cytoplasm, lamina propria uterus gland cell cytoplasm,and stromal cell cytoplasm,and it was lowly expressed in the beginning of the estrous cycle(P<0.05).Conclusion: Bcl-2,Bax, and Caspase-9 regulate the proliferation and apoptosis of endometrial cell cycle during estrous cycle
