ZPR1基因rs603446位点多态性与河南汉族人群血脂异常遗传易感性的关系

Abstract:
目的:研究河南汉族人群锌指蛋白(ZPR1)rs603446位点多态性与血脂异常的关联。方法:从河南农村队列研究中抽取916例血脂异常患者,根据年龄、性别进行1??GA6FA??1匹配,选取916例血脂正常者作为对照组; 采用2×48位SNPscanTM高通量SNP分型技术获取人群rs603446位点的基因型; 条件logistic回归分析位点多态性与血脂异常的关联强度; 多因子降维法(MDR)结合叉生分析评估基因与环境的交互作用。结果:血脂异常组和正常组ZPR1 rs603446位点3种基因型(CC、CT、TT)分布频率差异有统计学意义(P=0.046)。调整相关环境因素后,ZPR1基因rs603446位点突变基因型CT+TT携带者可降低血脂异常的发生(调整OR=0.695,95%CI=0.553~0.872); 交互作用分析结果显示,rs603446位点与吸烟具有相加交互作用(调整OR=0.516,95%CI=0.370~0.663),与体力活动具有相乘交互作用(调整OR=0.873,95%CI=0.819~0.930)。结论:ZPR1基因 rs603446多态性与血脂异常存在关联,且吸烟、体力活动对血脂异常的发病具有交互作用。
Aim: To investigate the association of zinc finger(ZPR1)gene rs603446 locus polymorphism and the susceptibility of dyslipidemia(DYS)in Henan Han population.Methods: A total of 916 DYS subjects and 916 controls based on 1:1 matching were collected from the Henan Rural Cohort study; the single nucleotide polymorphism of ZPR1 gene rs603446 was detected using a custom-by-design 2×48-Plex SNPscanTM Kit; conditional logistic regression was performed to evaluate the association between ZPR1 gene polymorphism and DYS; multifactor dimensionality reduction(MDR)model and crossover analysis were used to evaluate the interaction between gene and environment.Results: Three genotypes(CC, CT and TT)frequency of ZPR1 gene rs603446 between DYS group and control group had significant difference(P=0.046). The data showed that genotype(CT+TT)of ZPR1 gene rs603446 locus polymorphism decreased the risk of DYS after adjusting environmental risk factors(adjusted OR=0.695, 95%CI:0.553-0.872). Interaction analysis demonstrated that there was additive interaction between rs603446 and smoking(adjusted OR=0.516, 95%CI:0.370-0.663), and multiplicative interaction between rs603446 and physical activity on the susceptibility of DYS was found(adjusted OR=0.873, 95%CI:0.819-0.930).Conclusion: ZPR1 gene rs603446 locus polymorphism is associated with the susceptibility of DYS. The interaction between gene and smoking, gene and physical activity is found on the susceptibility of DYS
