Abstract:

目的:观察窖蛋白-1(Caveolin-1)在小鼠不同发育时期成釉细胞中的超微结构定位。方法:取胚胎发育第16.5天、第18.5天和出生第2.0天 的C57BL/6小鼠下颌第一磨牙牙胚,HE染色、透射电镜观察不同发育时期成釉细胞的组织学形态及超微结构变化,免疫电镜技术观察Caveolin-1在成釉细胞中的超微结构定位。结果:在各个发育阶段的成釉细胞细胞膜上均可见形态类似于胞膜窖的凹陷状结构,随着成釉细胞逐渐发育成熟,细胞器数量逐渐增多; 在不同发育阶段的成釉细胞中均可见Caveolin-1免疫阳性颗粒,在成熟成釉细胞的内质网腔中也可检测到Caveolin-1免疫阳性颗粒。结论:在小鼠下颌第一磨牙牙胚成釉细胞中,Caveolin-1定位于成釉细胞的细胞膜及成熟成釉细胞的内质网中。
Aim: To observe the location of Caveolin-1 in ameloblasts of mouse at different developmental stages.Methods: The lower first molar germs of C57BL/6 mice at embryonic day 16.5, 18.5 and the second postnatal day were collected. The histological morphology and ultrastructures of developing ameloblasts were investigated by HE staining and transmission electron microscope, respectively. The localization of Caveolin-1 protein was observed with immunoelectron microscopic techniques.Results: Caveolae-like invaginations were found on the membranes of ameloblasts at different developmental stages. Immunoelectron microscopy showed the depositions of Caveolin-1 positive electron particles on the membranes of developing ameloblasts. Caveolin-1 positive electron particles were also found in the chamber of endoplasmic reticulum(ER)in fully developed ameloblasts.Conclusion:Caveolin-1 locates on the cell membranes of mouse ameloblasts and in ER in fully developed ameloblasts
