黄芪多糖对HCT116细胞侵袭和迁移能力的影响

Abstract:
目的:研究黄芪多糖对人结直肠癌HCT116细胞侵袭和迁移能力的影响,并初步探讨其作用机制。方法:体外培养人结直肠癌HCT116细胞,分别以0.0、0.1、0.2,0.4、0.6、0.8和1.0 g/L的黄芪多糖干预HCT116细胞24 h,采用MTT法检测HCT116细胞的存活情况。采用0.0、0.1和0.6 g/L黄芪多糖作用HCT116细胞24 h后,Transwell和划痕实验检测侵袭和迁移能力,采用Western blot检测细胞中基质金属蛋白酶(MMP)-2、MMP-9蛋白表达水平。结果:黄芪多糖能够剂量依赖性抑制HCT116细胞增殖。0.6 g/L黄芪多糖处理HCT116细胞24 h后,细胞侵袭和迁移能力,MMP-2、MMP-9蛋白的表达水平均显著下降(P<0.05)。结论:黄芪多糖能够抑制HCT116细胞侵袭和迁移,其作用机制可能与下调MMP-2、MMP-9的表达有关。
Aim:To study the effects of astragalus polysaccharides(APS)on invasion and migration of human colorectal cancer HCT116 cells,and to explore its mechanism.Methods:HCT116 cells were cultured in vitro,and treated with different concentrations(0.0,0.1,0.2,0.4,0.6,0.8 and 1.0 g/L)of APS.The growth ability of HCT116 cells was detected by MTT assay after 24 h treatment,and the concentration of APS in the subsequent experiments was screened.HCT116 cells were treated with 0.0,0.1 and 0.6 g/L APS for 24 h,then Transwell and scratch test was used to detect the invasion and migration of HCT116 cells.The expression levels of matrix metalloproteinase-2(MMP-2)and matrix metalloproteinase-9(MMP-9)in HCT116 cells were detected by Western blot.Results:APS could inhibit the proliferation of HCT116 cells in a dose-dependent manner.The results of Transwell and scratch test showed that the invasiveness and migration ability of HCT116 cells treated with 0.6 g/L APS was significantly lower than that of the control group(P<0.05); APS could significantly inhibit the expression levels of MMP-2 and MMP-9 protein(P<0.05).Conclusion:APS can inhibit the invasion and migration of HCT116 cells,which may be related to the down-regulation of MMP-2 and MMP-9 expression
