精氨酸联合加味大柴胡汤对急性胆管炎大鼠肝脏功能的保护作用

Abstract:
目的:探讨加味大柴胡汤联合精氨酸对急性胆管炎大鼠肝脏功能的保护作用。方法:90只大鼠分为对照组、西药组、中药组、联合组和假手术组,每组18只。前4组建立胆道急性感染模型后,对照组给予生理盐水灌胃、西药组给予300 mg/kg精氨酸、中药组给予加味大柴胡汤(2 mL/次,2 次/d)灌胃、联合组同上给予精氨酸和加味大柴胡汤; 假手术组给予生理盐水灌胃。分别于处理后24、48及72 h取6只大鼠,取血测血浆内毒素与血清TB; 同时取肝组织,分别采用RT-PCR及免疫组化法检测TLR4 mRNA及蛋白的表达。结果:与假手术组相比,处理后24、48及72 h,对照组、西药组、中药组和联合组造模后血清TB水平、血浆内毒素水平及肝组织TLR4 mRNA表达水平升高,TLR4蛋白的表达增强; 与对照组相比,西药组、中药组及联合组以上指标均降低,其中联合组以上各指标最低(P<0.05)。结论:精氨酸联合加味大柴胡汤可能通过下调TLR4来降低内毒素水平从而减轻急性胆管炎大鼠的肝脏损害。
Aim: To explore the influence and mechanism of modified major decoction of bupleurum(MMDB)and arginine(Arg)on liver function of rats with acute cholangitis.Methods: A total of 90 rats were allocated into control group, western medicine group, medical herbs group, western medicine plus medical herbs group, and sham operation group(18 rats in each group). The first 4 groups were made acute cholangitis model. The rats in control group, western medicine group, medical herbs group, western medicine plus medical herbs group were given normal saline, Arg at 300 mg/kg,MMDB(2 mL per time and twice per day), and Arg plus MMDB,respectively. At 24,48 and 72 h after the above treatment, 6 rats were taken and the blood samples were prepared for determining plasma endotoxin and serum TB; the liver samples were prepared for detecting the expressions of TLR mRNA and protein by RT-PCR and immunohistochemical method,respectively.Results: Compared with sham operation group, serum TB and plasma endotoxin and the expressions of TLR mRNA and protein in control group, western medicine group, medical herbs group, western medicine plus medical herbs group were increased; compared with control group, the above indexes in western medicine group, medical herbs group, and western medicine plus medical herbs group were decreased, among which, those in western medicine plus medical herbs group were the lowest(P<0.05).Conclusion: MMDB plus Arg could reduce the level of plasma endotoxin by down-regulating the level of TLR4, thus alleviate the liver injury in rats with acute cholangitis
