Effects of puerarin on expressions of MMP？？2 and collagen Ⅳ mRNA after vascular injury in rats

目的：研究葛根素对大鼠颈动脉球囊损伤后新生血管内膜内基质金属蛋白酶？？2（MMP？？2）和Ⅳ型胶原（COL？并簦？mRNA表达的影响，进一步探讨其对血管重塑的干预作用。方法：将48只SD大鼠随机分为正常组、模型组和葛根素低、高剂量组，每组各12只。模型组和葛根素组均建立颈动脉球囊损伤模型；葛根素组术前7 d开始给予葛根素灌胃至实验结束，以生理盐水给模型组灌胃。各组分别于术后第14和28天处死6只大鼠，取动脉损伤段，HE染色观察血管形态学变化，采用RT？？PCR法检测动脉内MMP？？2和COL？并？ mRNA的表达。结果：术后14和28 d，模型组大鼠颈总动脉血管内膜比正常组增厚，用葛根素干预后新生内膜增生有所减轻；与正常组相比，模型组MMP？？2和COL？并？ mRNA表达增高(P＜0.05)；与模型组相比，葛根素组MMP？？2 mRNA表达下降，COL？并？ mRNA表达升高(P＜0.05)。结论：葛根素在大鼠颈动脉内皮细胞损伤后可能是通过下调MMP？？2和增强COL？并？ mRNA的表达，抑制平滑肌细胞从中膜到内膜的迁移，从而减轻新生内膜的增生。
Aim: To observe the effects of puerarin on the expressions of matrix metalloproteinase？？2(MMP？？2) and collagen Ⅳ(COL？并？) mRNA in rat carotid artery injured by balloon and to evaluate the mechanisms of inhibiting intima hyperplasia and vascular remodeling. Methods: The experimental model of carotid artery balloon injury in rats was established by endothelial injury induced by a balloon. Forty？？eight rats were randomly divided into 4 groups: normal group，model group, puerarin low dose group, and puerarin high dose group, and the latter 3 groups were made carotid artery balloon injury model. Rats of puerarin groups were treated with drugs for 2 and 4 weeks. Then the expressions of MMP？？2 and COL？并？ mRNA were assessed by RT？？PCR. Results: The hyperplasia of intima after balloon injury in the puerarin group was significantly milder than that of the model group. Compared with model group，the expression of MMP？？2 mRNA was significantly decreased， but the expression of COL？并？ mRNA was increased in the puerarin group(P<0.05). Conclusion: Puerarin could inhibit neointimal proliferation of rat carotid artery balloon injury，which may be explained by puerarin′s decreasing the level of MMP？？2 and increasing the level of COL？并？
