Abstract:

目的:探讨葛根素对多发性骨髓瘤细胞增殖和凋亡的影响及其机制。方法:取对数生长期的H929细胞,经0、20、40和80 μmol/L葛根素处理48 h,采用CCK-8法检测细胞增殖情况,流式细胞仪检测细胞的凋亡情况,Western blot检测细胞中AKT、p-AKT、Bcl-2和Cleaved Caspase-3蛋白的表达情况。结果:葛根素浓度依赖性地抑制H929细胞增殖,降低p-AKT和Bcl-2蛋白表达,诱导细胞凋亡,提高Cleaved Caspase-3蛋白表达(P<0.05)。结论:葛根素能够抑制H929细胞增殖并诱导细胞凋亡,其作用机制可能与抑制PI3K/AKT信号通路有关。
Aim:To investigate the effects of puerarin on proliferation and apoptosis of multiple myeloma H929 cells and its mechanism.Methods:H929 cells in logarithmic growth phase were treated with 0, 20, 40 and 80 μmol/L puerarin for 48 h.The proliferation of cells was detected by CCK-8 assay, apoptosis was detected by FCM, and the expressions of AKT, p-AKT, Bcl-2 and Cleaved caspase-3 protein in cells were detected by Western blot.Results:Puerarin inhibited the proliferation of H929 cells and the expressions of p-AKT and Bcl-2 protein, and induced apoptosis and increased Cleaved Caspase-3 protein expression in a concentration dependent manner(P<0.05).Conclusion:Puerarin can inhibit H929 cell proliferation,induce cell apoptosis,and the mechanism may be related to the inhibition of the PI3K/AKT signaling pathway
