THBS1表达上调对肺腺癌细胞增殖、凋亡和细胞周期的影响

Abstract:
目的:探讨血小板凝血酶蛋白1(THBS1)表达上调对肺腺癌细胞增殖、凋亡和细胞周期的影响。方法:采用实时荧光定量PCR检测肺腺癌细胞株A549、H1299、H358、H1975、PC9、PG49和Calu3中THBS1 mRNA的表达量; 构建真核表达载体pcDNA3.1-THBS1,以脂质体转染至THBS1 mRNA表达量最低的H1975细胞中,实时荧光定量PCR检测THBS1 mRNA表达水平,CCK-8法检测细胞增殖能力,流式细胞技术分析细胞凋亡及细胞周期分布情况。以未转染和转染空质粒pcDNA3.1的H1975细胞作对照。结果:不同肺腺癌细胞株中THBS1 mRNA表达量不同,H1975细胞中表达最低。与未转染和转染空质粒的H1975细胞比较,转染pcDNA3.1-THBS1的H1975细胞THBS1 mRNA表达上调(P<0.05),细胞增殖能力增强(P<0.05),细胞凋亡率降低(P<0.05),细胞周期进程被促进(P<0.05)。结论:THBS1与肺腺癌细胞的生物学行为关系密切,有可能成为肺腺癌的治疗靶点。
Aim:To investigate the effects of thrombospondin 1(THBS1)expression up-regulation on proliferation, apoptosis and cell cycle of lung adenocarcinoma cells.Methods:The expression levels of THBS1 mRNA in lung adenocarcinoma cell lines A549, H1299, H358, H1975, PC9, PG49 and Calu3 were detected by real-time quantitative PCR(qRT-PCR). Eukaryotic expression vector pcDNA3.1-THBS1 was constructed and transfected into H1975 cells by liposomes.qRT-PCR was used to detect the expression of THBS1 mRNA, CCK-8 kit was used to detect the proliferation,and flow cytometry was used to investigate the apoptosis and cell cycle distribution. The H1975 cells without transfection or transfected with pcDNA3.1 were the controls.Results:The expression levels of THBS1 mRNA in different lung adenocarcinoma cell lines were different, and the lowest was in H1975 cell line.Compared with the controls,the expression of THBS1 mRNA in H1975 cells transfected with pcDNA3.1-THBS1 was higher,proliferation ability was increased,the apoptosis rate decreased,and the cell cycle progression was promoted significantly(P<0.05).Conclusion:THBS1 protein is related with the biological behavior of lung adenocarcinoma cells, and may be a treatment target
