Abstract:

目的:研究雷公藤红素对人胃癌细胞HGC-27增殖、迁移及凋亡的影响。方法:用0.0、0.5、1.0、1.5、2.0、3.0、4.0 μmol/L的雷公藤红素培养HGC-27细胞24、48、72 h后,MTT法检测细胞增殖比率。用0.0、0.5、1.0、1.5 μmol/L的雷公藤红素培养HGC-27细胞48 h后,采用细胞划痕实验检测细胞迁移能力,流式细胞术检测细胞凋亡,Hochest染色检测细胞核凋亡小体的形成,Western blot法检测细胞中凋亡及侵袭相关蛋白Bcl-2、Bax、Cleaved Caspase-3、ERK、p-ERK、P38及p-P38的表达水平。结果:雷公藤红素作用后,HGC-27细胞增殖比率降低,细胞迁移率减小,早期细胞凋亡率增加(P<0.05)并能够诱导核凋亡小体的形成; 细胞中Bax、Cleaved Caspase-3、p-P38表达均升高,而Bcl-2和p-ERK表达降低(P<0.05)。结论:雷公藤红素对人胃癌细胞的增殖和迁移有一定的抑制作用,并能诱导其凋亡。
Aim: To investigate the effect of celastrol on proliferation, migration and apoptosis of human gastric cancer cell line HGC-27.Methods: MTT assay was used to detect the migration ability of HGC-27 cells treated with celastrol(0.0,0.5,1.0,1.5,2.0,3.0,4.0 μmol/L)for 24,48,72 h,respectively. The HGC-27 cells treated with celastrol(0.0,0.5,1.0,1.5 μmol/L)for 48 h were to detect the migration ability by scratch test, apoptosis by flow cytometry, nuclear apoptotic bodies by Hochest staining, apoptosis and invasion related proteins Bcl-2, Bax, Cleaved Caspase-3, ERK, p-ERK, P38, and p-P38 by Western blot.Results: The proliferation ratio of HGC-27 treated with celastrol decreased significantly, migration rate decreased,the early apoptotic rate increased(P<0.05)and nuclear apoptotic bodies formation was observed. The expressions of Bax,Cleaved Caspase-3 and p-P38 increased, while the expressions of Bcl-2 and p-ERK decreased(P<0.05).Conclusion: Celastrol can inhibit the proliferation and migration of human gastric cancer cells and induce apoptosis
