阿尔茨海默病患者认知功能及血清铜、同型半胱氨酸、尿酸水平、tau和Aβ1??42蛋白的检测*

Abstract:
目的:探讨血清铜、同型半胱氨酸(Hcy)、尿酸(UA)水平、tau和Aβ1??42蛋白与阿尔茨海默病(AD)患者认知功能及生活质量的关系。方法:分别采用等离子体原子吸收光谱法检测96例AD患者及120例健康对照者血清铜水平,循环酶法测定血清Hcy水平,比色法测定血UA水平,ELISA试验检测血清中tau和Aβ1??42蛋白水平,采用简易智能量表(MMSE)评价认知功能,采用世界卫生组织生活质量测定简表(中文版)评价生活质量。结果:与对照组相比,AD组血清铜(t=4.744,P<0.001)、Hcy(t=7.174,P<0.001)和tau蛋白(t=4.238,P<0.001)水平明显升高,UA(t=8.225,P<0.001)、Aβ1??42蛋白水平(t=8.781,P<0.001)明显降低。Pearson相关分析显示AD患者MMSE评分与血清铜、Hcy水平呈负相关(r=-0.450、-0.537;P<0.001),与UA、Aβ1??42无关;生活质量评分与血清铜、Hcy水平呈负相关(r=-0.490、-0.550;P<0.001),与UA、Aβ1??42无关。结论:血清铜、Hcy水平与AD患者认知功能、生活质量有关,UA可能在AD的防治中起积极作用。
To study the relationship between serum copper, Hcy, uric acid(UA), tau and Aβ1??42 protein levels and cognitive function, the life quality of patients with Alzheimer disease(AD).Methods: A total of 96 patients with AD and 120 normal healthy persons were selected to measure the level of serum copper by atomic emission spectrometry, the level of serum Hcy by enzymatic cycling assay, and the level of serum UA by in vitro enzymatic colorimetric test; Aβ1??42 and tau proteins were analyzed by ELISA; using mini??mental state examination(MMSE) to evaluate the cognitive function, using the Chinese version of WHO quality of life scale??brief form questionnaire(WHOQOL??BREF) to evaluate the quality of life. Results: Compared with the normal control group,the level of serum copper(t=4.744,P<0.001), Hcy level(t=7.174,P<0.001) and tau protein(t=4.238,P<0.001) in AD patients were significantly higher; the serum UA level was significantly lower(t=8.225,P<0.001), the serum Aβ1??42 level was significantly lower(t=8.781,P<0.001).The data based on AD patients showed negative correlation between serum copper level,serum Hcy level and MMSE(r=-0.450,-0.537;P<0.001); there was negative correlation between serum copper, serum Hcy level and WHOQOL??BREF(r=-0.490,-0.550;P<0.001). Conclusion: The serum copper and Hcy levels are associated with the cognitive impairment and life quality in patients with AD, and the serum UA level may play a protective role against AD
