Abstract:

摘要:目的 检测IL-1β和IL-4在湿热证溃疡性结肠炎(UC)大鼠血清中的表达,探索其在评价炎症严重程度中的作用及意义。方法 将50只健康SPF级Wistar大鼠(雌雄各半),随机分成5组:正常组及湿热证UC模型Ⅰ(3d)组、Ⅱ(7d)组、Ⅲ(14d)组、Ⅳ(21d)组,每组均10只大鼠。除正常组在自然环境饲养外,其余各湿热证UC模型组均置于高脂高糖饮食及高温高湿环境中,喂养4周后,用TNBS-乙醇灌肠法诱发湿热证UC。检测各组大鼠疾病活动指数(DAI)、结肠黏膜损伤指数(CMDI)、IL-1β及IL-4含量的变化。结果 与正常组比较,湿热证UC模型Ⅰ、Ⅱ、Ⅲ组大鼠DAI、CMDI、IL-1β含量显著升高(P<0.01、P<0.05),模型Ⅱ组升高最为明显(P<0.01);血清IL-4含量明显下降(P<0.01、P<0.05),模型Ⅱ组下降最为明显(P<0.01);模型Ⅳ组上述指标均无统计学差异(P>0.05)。结论 在湿热型UC发病过程中炎性细胞因子IL-1β、IL-4参与了其发病过程,且IL-1β上升及IL-4下降水平同炎症严重程度在时间上是一致的,可作为炎症严重程度的评价指标。
ABSTRACT: Objective To study the expressions of serum interleukin-1β (IL-1β) and interleukin-4 (IL-4) in ulcerative colitis (UC) rats with damp-heat syndrome and explore their significance in assessing inflammatory severity. Methods We randomly divided 50 healthy Wistar rats of SPF grade (half males and half females) into normal group, and damp-heat syndrome model of UC Ⅰ (3 days), Ⅱ (7 days), Ⅲ (14 days) and Ⅳ (21 days) groups, with 10 in each group. Rats in normal group were raised in the natural environment of breeding, while those UC model groups with damp-heat syndrome were fed with high-sugar and high-fat food in the high-temperature and high-humidity environment for 4 weeks. The rat model of damp heat syndrome UC was established by using TNBS-ethanol enema. Then we detected the changes of disease activity index (DAI), colonic mucosa damage index (CMDI), IL-1β and IL-4 in each group. Results Compared with those in normal control group, DAI, CMDI and IL-1β levels in damp heat syndrome UC model Ⅰ, Ⅱ, Ⅲ groups were significantly increased (P<0.01, P<0.05), with model group Ⅱ having the most obvious increase (P<0.01). Serum IL-4 level decreased significantly (P<0.01, P<0.05), with model group Ⅱ having the most significant decrease (P<0.01). The above indexes did not significantly differ in model group Ⅳ (P>0.05). Conclusion Inflammatory cytokines IL-1β and IL-4 are involved in the pathogenesis of damp-heat syndrome UC. The increased IL-1β and decreased IL-4 levels are consistent with the time of inflammatory severity; therefore, they can be regarded as markers for assessing inflammatory severity in UC
