NRP-1在胃癌组织及细胞系中的表达及临床意义

Abstract:
摘要:目的 研究神经纤毛蛋白1(NRP-1)在胃癌组织的表达,探讨其与胃癌发生、发展及临床病理因素的关系及可能的临床意义。方法 应用免疫组织化学染色检测180例胃癌患者的胃癌组织及癌旁组织标本NRP-1表达,用免疫印迹方法检测胃癌组织及不同分化程度的胃癌细胞NRP-1表达,K-M生存曲线和多因素回归模型用于分析胃癌预后。结果 胃癌组织中NRP-1表达水平明显高于癌旁组织(P<0.001),胃癌组织中NRP-1高表达与肿瘤分化程度、淋巴结转移、AJCC分期、T分期呈正相关(P<0.05)。多因素生存分析表明高表达NRP-1是胃癌患者总生存期的独立预后因子。 结论 过表达NRP-1可能在胃癌进展中发挥着重要作用,过表达NRP-1可作为生物标志物预测胃癌患者的不良预后。
ABSTRACT: Objective To analyze the expression of neuropilin-1 (NRP-1) in gastric cancer tissues and assess its clinicopathological parameters and prognostic significance. Methods NRP-1 expression was determined by immunohistochemistry in specimens of primary cancer tissues and the adjacent non-cancerous tissues in 180 patients with gastric cancer. NRP-1 expression was analyzed by Western blot in gastric cancer tissues and different gastric cancer cell line. Kaplan-Meier plots and Cox’s proportional hazards regression model were used to analyze the prognosis of gastric cancer. Results By immunohistochemistry, the expression of NRP-1 was significantly higher in gastric cancer than that in adjacent noncancerous tissues (P<0.001). High expression of NRP-1 was positively associated with histologic grade, tumor stage, lymph node metastasis and tumor T stage (P<0.05). Multivariate analysis showed that the high expression of NRP-1 was an independent prognostic factor for overall survival. Conclusion Overexpression of NRP-1 may play an important role in the progression of gastric cancer, and NRP-1 overexpression may serve as a biomarker for poor prognosis of gastric cancer patients
