新疆哈萨克族原发性高血压与KCNMB1基因多态性的关系

Abstract:

摘要:目的 探讨新疆哈萨克族原发性高血压与大电导钙激活钾通道(large-conductance Ca2+-activated potassium channel, BKCa)KCNMB1基因多态性位点rs11739136的相关性。方法 分别提取原发性高血压组(essential hypertension group, EH, n=277)和正常组(normotensive group, NT, n=247)血液中的基因组DNA,运用限制性片段长度多态性聚合酶链式反应(polymerase chain reaction-restriction fragment length polymorphism, PCR-RFLP)技术检测KCNMB1-rs11739136多态性位点的基因型。应用非条件Logistic回归分析新疆哈萨克族高血压相关危险因素。结果 非条件Logistic 回归分析结果显示,KCNMB1-rs11739136位点基因型、高密度脂蛋白(high-density lipoprotein, HDL)、低密度脂蛋白(low-density lipoprotein, LDL)和总胆固醇等与高血压无关联;体质量指数(body mass index, BMI)和甘油三酯是影响高血压的危险因素。新疆哈萨克族人群中普遍存在KCNMB1-rs11739136位点的基因多态性(P>0.05)。在EH组该位点CC、CT、TT基因型频率及C、T等位基因频率分别为74.00%、23.83%、2.17%、85.92%、14.08%,与NT组相比较,两组间无统计学差异(P>0.05)。结论 KCNMB1基因rs11739136多态性位点可能与新疆哈萨克族原发性高血压无关联。
ABSTRACT: Objective To explore the correlation between the single nucleotide polymorphism of large-conductance Ca2+-activated potassium channel (BKCa)-rs11739136 and essential hypertension in Xinjiang Kazak population. Methods Genomic DNA was extracted from the blood of essential hypertension group (EH group, n=277) and normal group (NT group, n=247), respectively. The polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to detect genotypes of rs11739136 polymorphism of KCNMB1. The unconditional logistic regression analysis was used to evaluate the risk factors associated with Xinjiang Kazak hypertension. Results The unconditional logistic regression analysis showed that KCNMB1-rs11739136 genotypes, high-density lipoprotein ( HDL), low-density lipoprotein (LDL) and cholesterol were not associated with hypertension, while body mass index (BMI) and triglyceride were risk factors for hypertension. KCNMB-rs11739136 polymorphism existed extensively in Xinjiang Kazakh population (P>0.05). The frequencies of CC, CT, TT genotypes and those of alleles C and T were 74.00%, 23.83%, 2.17%, 85.92%, 14.08% in EH group, respectively. NT and EH groups did not significantly differ (P>0.05). Conclusion The rs11739136 polymorphism of KCNMB1 may not be related to essential hypertension in Xinjiang Kazak population
