Abstract:

摘要:目的 探讨舒芬太尼对肝癌Hep3B细胞活性的影响及相关机制。方法 以Hep3B肝癌细胞为研究对象,经不同浓度(0.01、0.1、1、5、10、15μmol/L)舒芬太尼处理后,MTT法观察细胞活性的改变,流式细胞仪及AnnexinV/PI双标记染色检测Hep3B细胞周期及凋亡的变化,Western blot检测survivin及caspase-3蛋白的表达。结果 随着舒芬太尼浓度的增加,肝癌细胞活性降低,细胞周期停滞在G0/G1期,细胞凋亡增多,survivin蛋白表达量降低,caspase-3蛋白表达量升高。结论 舒芬太尼能通过改变肝癌细胞的细胞周期,促进细胞凋亡,改变survivin和caspase-3蛋白的表达,抑制Hep3B细胞的活性。
ABSTRACT: Objective To investigate the effects and mechanism of Sufentanil on Hep3B cell viability in liver cancer. Methods Sufentanil of different concentrations (0.01, 0.1, 1, 5, 10, 15μmol/L) was used to treat Hep3B cells. The changes of cell cycle, apoptosis and protein expression were detected to explore the potential mechanisms by MTT method, flow cytometry and Western blot, respectively. Results Reduced viability of Hep3B cells, arrested cell cycle in the G0/G1 phase, decreased expression of survivin protein, and increased expression of caspase-3 protein were observed with the increase of Sufentanil concentration. Conclusion Sufentanil inhibited the viability of Hep3B cells through affecting cell cycle, promoting cell apoptosis and changing protein expressions of survivin and caspase-3
