针灸对实验性糖尿病小鼠心脏功能调节的影响

Abstract:

摘要:目的 探讨电针与艾灸足三里穴对糖尿病小鼠心功能及运动功能的影响及其治疗效应是否存在差异。方法 8~9周龄C57雄性小鼠一次性链脲佐菌素(STZ)腹腔注射,建立1型糖尿病动物模型;空白组腹腔注射等量的枸橼酸钠缓冲液。造模3周后,将糖尿病小鼠再随机分为3组:艾灸干预组、电针干预组和糖尿病组。治疗前后测量各组小鼠血糖和体质量;治疗8周后,在11周末彩色超声心动图检测心脏结构与功能,包括左心室收缩末内径(LVESd)、缩短分数(FS);滚轴测定运动能力,包括坠落时间(潜伏期)和爬行距离。结果 糖尿病组超声心动图短轴缩短率较空白对照组明显降低(P<0.05);电针干预组、艾灸干预组治疗后超声心动图短轴缩短率较空白组无统计学差异(P>0.05)。糖尿病组滚轴实验体能测试坠落潜伏期较空白组显著降低(P<0.05);电针干预组、艾灸干预组治疗后,滚轴实验坠落潜伏期较空白组均无统计学差异(P>0.05)。电针干预组相比艾灸干预组心功能无明显变化(P>0.05)。结论 STZ注射后11周,小鼠心脏收缩功能显著降低,运动能力明显下降,电针和艾灸足三里均能改善STZ诱导的小鼠左心室收缩功能,且两种治疗方法效应相当。
ABSTRACT: Objective To investigate the effects of electroacupuncture and moxibustion at Zusanli point on cardiac function and motor function in diabetic mice and whether there is any difference in their therapeutic effects. Methods Male C57 mice aged 8 weeks were injected intraperitoneally with streptozotocin STZ to establish the animal model of diabetic cardiomyopathy. The control group was injected intraperitoneally with the same amount of sodium citrate buffer solution. After 3 weeks of modeling, diabetic mice were tested for blood glucose and body weight. They were randomly divided into 3 groups: moxibustion intervention group, electroacupuncture intervention group and diabetes group. After 8 weeks of treatment, the cardiac structure and function including left ventricular end-systolic diameter (LVESd) and shortening score (FS) were measured by color echocardiography at 11th weeks. The use of roller determined exercise capacity, including the fall time (latency) and crawling distance. Results The shortening rate of short axis of echocardiography in diabetic group was significantly lower than that in blank group (P<0.05). There was no difference in echocardiographic short axis shortening rate between electroacupuncture and moxibustion intervention groups (P>0.05). The fall latency of roller test was significantly lower in diabetic group than in blank group (P<0.05). After treatment electroacupuncture intervention group and moxibustion intervention group did not differ from blank group in fall latency of roller test (P>0.05). Cardiac function did not change significantly in electrocardiogram intervention group compared with moxibustion intervention group (P>0.05). Conclusion Diabetic mice showed significantly decreased cardiac function and exercise capacity 11 weeks after STZ injection. Electroacupuncture and moxibustion at Zusanli can improve left ventricular systolic function. However, there is no difference in the therapeutic effect between the two treatments
