百草枯染毒大鼠支气管肺泡灌洗液中细胞因子的变化

Abstract:

目的探讨百草枯(PQ)中毒大鼠早期支气管肺泡灌洗液(BALF)中细胞炎性因子的变化。 方法将成年健康SD大鼠随机分为正常对照组(n=8)、百草枯一次性灌胃染毒低剂量组(PQ 15 mg/kg,n=24)、中剂量组(PQ 30 mg/kg,n=24)、高剂量组(PQ 60 mg/kg,n=24)。对照组给予相同容积生理盐水灌胃。每组分别于染毒后第1、7、14、21 d处死大鼠,收集BALF。采用ELISA法测定白细胞介素1(IL-1)、IL-6、巨噬细胞炎性蛋白2(MIP-2)、单核细胞趋化因子1(MCP-1)、新喋呤的浓度。 结果各组BALF中炎性细胞因子在各时间点较对照组均有不同程度的增高。在染毒1~14 d时BLAF中IL-1、新喋呤浓度随染毒剂量的增加而增高趋势明显,剂量反应关系明显,差异有统计学意义(P<0.05)。14、21 d时IL-6随染毒剂量增加而增高的趋势较明显。染毒组IL-l、IL-6、新喋呤均在14 d时出现浓度峰值。高剂量组14 d时MIP-2明显高于低、中剂量组;低剂量组各时间点MCP-1明显低于中、高剂量组,差异均有统计学意义(P<0.05)。 结论炎症因子在PQ染毒后炎症发生发展过程中发挥了重要作用,可能促进肺损伤的发生与发展
