抗体免疫脂质磁球法检测肺癌循环肿瘤细胞初探

Abstract:

目的 初步探索抗体免疫脂质磁球法在肺癌循环肿瘤细胞(CTCs)检测中的应用。 方法 采用反向蒸发法制备上皮细胞粘附分子(EpCAM)和表皮生长因子受体(EGFR)抗体免疫脂质磁球。在磷酸盐缓冲溶液(PBS)和外周血肺癌CTCs模型中,评价EpCAM和EGFR抗体免疫脂质磁球对A549细胞的捕获效率;在此基础上,用上述两种抗体免疫脂质磁球法检测肺癌患者外周血中CTCs;并将EpCAM免疫脂质磁球法与CellsearchTM系统进行比较。 结果 EpCAM和EGFR免疫脂质磁球法在PBS溶液CTCs模型中对A549细胞的中位检出率分别为92%和90%,在外周血模型中分别为85%和81%;两种磁球法在13例肺癌患者中均检测出了CTCs,中位检出个数均为5个,而在健康对照者中中位检出个数均为0个(P<0.05)。用EpCAM免疫脂质磁球法和CellsearchTM系统检测3例肺癌患者的CTCs,前者对其中2例患者的CTCs检出量高于后者,另外1例相等。 结论 EpCAM和EGFR免疫脂质磁球法均能有效检出肺癌CTCs,两种磁球法检出率相当。EpCAM免疫脂质磁球法对肺癌患者CTCs检出量有高于CellsearchTM系统的趋势
