Sonic Hedgehog信号通路在成年大鼠脊髓损伤后的表达

Abstract:

目的观察Sonic Hedgehog(Shh)信号通路成分在成年大鼠脊髓损伤(spinal cord injury,SCI)后是否表达,探讨Shh信号通路的表达规律及意义。 方法取健康雄性SD大鼠64只,随机分为正常组(A组,8只)、假手术组(B组,8只)和SCI组(C组,48只)。A组大鼠不作任何处理;B组大鼠仅咬开T7~9椎板,不作损伤处理;C组大鼠采用改良Allen法制作大鼠SCI模型。取A、B组及C组术后12 h,1、3、7、14、21 d大鼠(n=8),采用BBB评分标准评估大鼠后肢运动恢复情况,免疫荧光染色、实时荧光定量PCR及Western blot法检测SCI区Shh、Gli-1(Glioma-associated oncogene homolog-1)mRNA和蛋白表达水平变化。 结果C组BBB评分随时间延长缓慢升高,但各时间点BBB评分均显著低于A、B组(P<0.05)。免疫荧光染色示,C组SCI后可见Shh、Gli-1大量增生。实时荧光定量PCR和Western blot结果示,C组SCI后Shh、Gli-1 mRNA相对表达量和蛋白表达量均逐渐增高,7 d时达最大值,各时间点Shh、Gli-1 mRNA相对表达量和蛋白表达量均显著高于A、B组(P<0.05)。相对于A组,C组SCI后Gli-1蛋白在细胞质中表达减少,细胞核中表达增加,呈现出“核转移”现象。 结论SCI后星形胶质细胞可分泌Shh、Gli-1信号分子,两者表达均显著上调并表现出规律的动态变化,提示Shh信号通路可能参与了SCI后神经细胞再生修复的调控
