基于MRI数据的3D打印定制脑组织空腔支架的研究

Abstract:

目的探讨3D打印技术制备个性化大鼠脑组织空腔支架方法,为组织工程化定制空腔支架进行创伤性颅脑损伤(traumatic brain injury,TBI)修复奠定基础。 方法取成年雄性SD大鼠5只,体质量(300±10)g,使用电子控制性脑皮质撞击仪建立TBI模型,基于MRI数据采用Mimics软件重建损伤空腔表面轮廓,计算机辅助设计构建内部结构,再用3D生物打印机以胶原-壳聚糖复合物为原料进行3D打印。对3D打印支架进行外观及扫描电镜观察。 结果MRI扫描显示大鼠致伤侧脑组织损伤改变,且空腔位置均限于大脑右侧皮层区。成功构建了含有内部结构的个性化空腔三维模型,并利用3D打印技术成功制备了空腔支架。支架外部轮廓与大鼠TBI后损伤区空腔相似,内部正交叉结构排列有序、孔隙连通性较好。 结论结合基于MRI数据的三维重建及3D打印技术定制的空腔支架与大鼠脑组织缺损空腔外形相近,内部正交叉结构可模拟细胞外基质拓扑结构,且打印材料为生物相容性较好的胶原-壳聚糖复合物,为定制空腔支架修复TBI后脑组织空腔的研究提供了一种新方法
