取食不同水稻品种对褐飞虱和稻株内海藻糖及海藻糖酶的影响

Abstract:

为明确不同水稻品种与褐飞虱的相互作用关系,以水稻品种TN1(感虫品种)、中浙优1号(生产主栽品种)和IR56(抗虫品种)为研究对象,分别采用蒽酮法和葡萄糖分析试剂测定了褐飞虱取食不同水稻品种后对褐飞虱和稻株体内的海藻糖含量及海藻糖酶活性的影响。结果显示,被褐飞虱取食后,IR56和中浙优1号的海藻糖酶活性分别由虫状态下的3.56、4.34 nmol·g-1·min-1显著升高至7.53、6.05 nmol·g-1·min-1,海藻糖含量分别由12.10、5.14 nmol/μg降至3.14、2.43 nmol/μg,而TN1的海藻糖酶活性由1.46 nmol/μg显著升高至3.05 nmol/μg,海藻糖含量明显变化。取食TN1、IR56和中浙优1号后,褐飞虱体内的海藻糖含量分别为0.53、0.28和0.20 nmol/μg,取食后二者的褐飞虱体内海藻糖含量与TN1组相比较均显著降低;且褐飞虱体内海藻糖酶活性以及3个海藻糖酶基因在mRNA水平均显著低于TN1组。表明海藻糖及海藻糖酶在褐飞虱与抗虫水稻的相互作用中可能发挥着重要的调控作用。
To clarify the interaction between different rice varieties and Nilaparvata lugens (St?l), TN1(susceptible variety), Zhongzheyou No.1(major variety in Zhejiang) and IR56(insect-resistant variety) were chosen to determine the effects of N. lugens infestation on the trehalose contents and trehalase activities in both rice and N. lugens by using anthrone-sulfuric acid method and glucose analytical reagent, respectively. The results showed that the trehalase activity was increased from 3.56 and 4.34 nmol·g-1·min-1 to 7.53 and 6.05 nmol·g-1·min-1 in IR56 and Zhongzheyou No.1 infested by N. lugens, and the trehalose content was decreased from 12.10 and 5.14 nmol/μg to 3.14 and 2.43 nmol/μg. While the trehalase activity was increased from 1.46 nmol/μg to 3.05 nmol/μg in TN1 group, the trehalose content was not significantly changed. The trehalose contents in N. lugens feeding on TN1, IR56 and Zhongzheyou No.1 were 0.53, 0.28 and 0.20 nmol/μg, respectively. Compared with TN1 group, the trehalose content in IR56 and Zhongzheyou No.1 groups were significantly lower, and the trehalase activity and expression of three trehalase genes at mRNA expressional level were also declined significantly. These results indicated that trehalose and trehalase play important roles in the interaction between insect-resistant rice and N. Lugens.
