Abstract:

摘要:目的:观察力竭运动应激后对小鼠脾脏T、B淋巴细胞钾离子通道表达的差异性,探讨力竭运动应激对T、B淋巴细胞的影响。方法:复制小鼠力竭模型后从脾脏中分离出T、B淋巴细胞,测定淋巴细胞转化功能,溶血空斑形成实验,流式细胞仪T淋巴细胞周期和分型,RT-PCR、Western blot测小鼠脾脏T、B细胞Kv1.3的表达。结果:小鼠脾脏T淋巴细胞Kv1.3 mRNA和蛋白质表达水平明显下调(P<0.05),小鼠脾脏B淋巴细胞Kv1.3 mRNA和蛋白质表达水平没有显著性差异(P>0.05);力竭运动应激小鼠T淋巴细胞功能受抑制(P<0.05),B淋巴细胞功能未受到明显影响(P>0.05)。结论:力竭运动应激导致小鼠脾脏T、B淋巴细胞Kv1.3 mRNA和蛋白质表达呈现差异性,进而影响淋巴细胞功能,提示力竭运动应激条件下细胞免疫功能的抑制可能与钾通道有关,为进一步研究运动应激导致免疫抑制的机制提供了新的思路。
Abstract: Objective: This study aimed at observing different expressions of Kv1.3 of T and B lymphocytes in spleen of mice after exhaustive exercise, and exploring the effect of exhaustive exercise on T and B lymphocytes. Methods: T and B lymphocytes in spleen of mice were separated after exhaustive exercise; lymphocyte transformation test(LTT), plague forming cell (PFC) test were performed to demonstrate their functions; RT-PCR, Western blot were performed to demonstrate the mRNA and protein expressions of Kv1.3 of T and B lymphocytes in spleen. Results: Compared with control group, the mRNA and protein expressions of Kv1.3 were significantly down-regulated in T lymphocyte of spleen after exhaustive exercise (P < 0.05), and there were no significant difference of B lymphocyte after exhaustive exercise (P < 0.05); transformation function of T lymphocyte was affected by exhausted exercise(P < 0.05) and that of B lymphocyte was not (P < 0.05). Conclusion: Exhaustive exercise changes the mRNA and protein expressions of Kv1.3 of T and B lymphocytes in spleen of mice, affects the function of lymphocytes, which indicate that the inhibition of cellular immune function under exhaustive exercise stress may be related to potassium channels. This study provided a new idea for further research on the mechanism of exercise inducing immunosuppression
