Abstract:

以紫菜为原料,通过乙醇溶剂提取、硅胶柱层析、高效液相制备色谱等步骤,得到紫菜多酚组分C1、C2、D1、D2四个组分. 通过MTT法测定组分C1、C2对人恶性黑色素瘤细胞A375的增殖抑制作用,结果表明:组分C1、C2作用于A375细胞48h的IC50分别为1.10、1.58mg·mL-1. 采用Annexin V/PI双染测定细胞凋亡情况发现,A375细胞经紫菜多酚组分C1处理后凋亡的细胞比例增加. 经组分C1作用的A375细胞中黑色素含量和相对酪氨酸酶活力均得到提高,且与药物浓度呈正相关关系;透射电镜检测A375细胞超微结构的变化发现紫菜多酚组分C1可诱导A375细胞趋向正常分化.
Used laver as raw material,the four monomer components C1,C2,D1,D2 of laver polyphenol were obtained by ethanol solvent extraction,silica gel column chromatography,and HPLC preparative chromatography. The MTT method was adopted to determine the proliferation inhibition effect of components C1,C2 on human malignant melanoma cell A375. The results showed that the IC50 of component C1,C2 on cell A375 for 48hours is 1.10,1.58mg·mL-1 respectively. The cell apoptosis was examined by the Annexin V/PI double staining. It was found that the proportion of apoptosis of cell A375 increases after the treatment of component C1 of laver polyphenol. The content of melanin and the relative activity of tyrosinase in cell A375 also increased after the treatment of component C1,positively correlated with drug concentration. Component C1 of laver polyphenols can induce cell A375 to normal differentiation through testing cell ultrastructure by transmission electron microscope
