Abstract:

摘要 目的: 探讨增强芳香烃受体(AhR)对高糖状态牙龈卟啉单胞菌(Pg)脂多糖(LPS)导致的巨噬细胞炎症反应的影响。方法: 将小鼠巨噬细胞RAW264.7接种在高糖培养基中,并转染特异性增强AhR表达的重组质粒,稳定转染后将细胞在高糖状态加入Pg LPS刺激。用免疫印迹实验检测细胞的AhR及炎症反应相关蛋白Notch1、Jagged1、iNOS的表达,并用ELISA检测细胞培养上清液中的促炎因子TNFα、IL1β浓度。结果: 在受高糖加Pg LPS刺激后,与未增强AhR表达的对照组细胞相比,增强AhR表达的细胞Notch1、Jagged1、iNOS、TNFα、IL1β蛋白表达量减少(P<0.05)。结论: 增强AhR表达,可抑制高糖状态Pg LPS导致的巨噬细胞炎症反应,该抑制作用可能与AhR调节Notch通路有关
