Abstract:

摘要 目的: 比较二维及三维方式培养的人舌鳞状细胞癌(简称鳞癌)TCA8113细胞株对细胞的增殖、组织形态及乳酸脱氢酶(lactate dehydrogenase,LDH)活性的影响,为体外舌鳞癌的研究提供培养方法。方法: 倒置显微镜和苏木精-伊红(hematoxylin and eosin,HE)染色观察二维及三维培养的人舌鳞癌TCA8113细胞的形态;噻唑蓝(methyl thiazolyl tetrazolium,MTT)法观察两种培养体系中细胞的增殖;酶联免疫吸附试验(Enzyme-linked immunosorbent assay,ELISA)检测两种培养体系中LDH活性的改变。结果: 倒置显微镜和HE染色观察到三维培养体系中TCA8113细胞成团生长;与二维培养体系比较,三维培养体系中细胞较长时间保持良好的增殖状态,LDH的活性较二维培养的细胞活性显著增高。结论: TCA8113细胞藻酸盐凝胶三维培养体系较二维培养体系能更好地体现细胞的生物学特性
