舌鳞状细胞癌淋巴结转移与长链非编码RNA HOTAIR的关系探讨

Abstract:
摘要 目的:研究HOTAIR在舌鳞状细胞癌中的表达,并探究HOTAIR与舌鳞状细胞癌淋巴结转移的关系。方法:对19例舌鳞状细胞癌患者的癌组织以及其对应的19例癌旁组织进行实时荧光定量PCR检测,检测HOTAIR在癌组织、癌旁组织的相对表达量,把结果与患者的TNM分期进行关联比较,并进一步通过构建沉默HOTAIR表达的CAL27、SCC9细胞系,检测肿瘤细胞侵袭、迁移能力的改变。结果:19对舌鳞状细胞癌组织中,26.3%显示出HOTAI的高表达(P=0.016 9<0.05),且高表达者均为具有淋巴结转移的患者,而在无淋巴结转移的癌组织中HOTAIR呈现低表达(P<0.001),通过RNA干扰技术降低HOTAIR在CAL27及SCC9细胞系中的表达水平,结果发现细胞侵袭(P<0.001)和迁移能力(P<0.001)受到了抑制。结论:长链非编码RNA HOTAIR在舌鳞状细胞癌淋巴结转移过程中可能发挥促进因子的作用
