Abstract:

摘要 目的:探究水溶性石墨烯转染滑膜间充质干细胞的的效果及各组间细胞克隆形成率的差别比较。方法:将传代培养到第3代的滑膜间充质干细胞分成实验组与对照组,实验组细胞培养基内加入浓度为25 mol/mL的水溶性石墨烯在37 ℃的条件下培养24 h,对照组用普通培养基培养37 ℃ 24 h,405 nm激发波长下的激光共聚胶显微镜下观察石墨烯是否成功转染滑膜干细胞。各组间细胞克隆形成率的差别采用单因素方差分析比较。结果:实验组的细胞培养24 h后,在激光共聚焦显微镜下细胞质可见发光的荧光点,细胞核的光强度较弱。各组克隆形成率均P<0.05有显著差异。结论:石墨烯容易渗透到细胞质但没有进入细胞核,可以猜测石墨烯在进入活的滑膜间充质干细胞后没有发生遗传的中断
