硫酸软骨素蛋白聚糖对MC3T3-E1细胞株矿化的影响

Abstract:
摘要 目的:观察硫酸软骨素蛋白聚糖(chondroitin sulfate proteoglycan,CSPG)对前成骨细胞株MC3T3-E1矿化过程中碱性磷酸酶(alkaline phosphatase,ALP)活性和钙离子沉积量的影响。方法:MC3T3-E1细胞株在含不同浓度CSPG(0、5、10、15 mg/L)诱导液中培养,不加CSPG组为对照组,其余3组为实验组,细胞诱导14 d时行ALP染色和活性检测,21 d时行茜素红染色和钙离子沉积量检测。结果:与对照组比较,各实验组ALP和茜素红染色面积随CSPG浓度增加逐渐减小,颜色变浅;定量分析结果显示,钙离子沉积量整体差异有统计学意义(P<0.05),ALP活性整体差异无统计学意义。结论:在本实验周期内,CSPG以剂量依赖方式抑制MC3T3-E1细胞株钙离子的沉积
