Abstract:

摘要 目的: 探究芦荟大黄素纳米脂质体(aloe emodin nano-liposome,AE-L)介导的光动力疗法(photodynamic therapy,PDT)对人舌鳞状细胞癌(简称鳞癌)细胞Tca-8113的治疗效果。方法: 逆相蒸发法制备AE-L;不同浓度AE-L(2.5、5.0、7.5、10.0、12.5 μg/mL)作用Tca-8113细胞4 h后,不同光照时间处理(0、10、20、40、80、160 s;激发波长为405 nm,功率密度为80 mW/cm2)Tca-8113,采用噻唑蓝(methyl thiazol tetrazolium,MTT)检测细胞存活率;AE-L(7.5 μg/mL)作用细胞4 h后,光照时间为80 s(激发波长为405 nm,功率密度为80 mW/cm2)处理细胞,荧光显微镜定性分析PDT后活性氧(reactive oxygen species,ROS)含量。结果: AE-L浓度≤7.5 μg/mL时细胞毒性较小,与对照组比较差异无统计学意义(P>0.05)。PDT组随着光照时间延长细胞存活率逐渐降低,光照时间80 s时细胞存活率为76.6%,与对照组比较差异有统计学意义(P<0.001)。荧光染色结果显示,PDT治疗后细胞内ROS含量显著增加。结论: 7.5 μg/mL AE-L介导的PDT治疗对人舌鳞癌Tca-8113细胞具有显著的杀伤效果
